RESUMO

Limited resources affect an organism's physiology through the conserved metabolic pathway, the mechanistic target of rapamycin (mTOR). Males and females often react differently to nutritional limitation, but whether it leads to differential mTOR pathway expression remains unknown. Recently, we found that dietary restriction (DR) induced significant changes in the expression of mTOR pathway genes in female Japanese quails (Coturnix japonica). We simultaneously exposed 32 male and female Japanese quails to either 20%, 30%, 40% restriction or ad libitum feeding for 14 days and determined the expression of six key genes of the mTOR pathway in the liver to investigate sex differences in the expression patterns. We found that DR significantly reduced body mass, albeit the effect was milder in males compared to females. We observed sex-specific liver gene expression. DR downregulated mTOR expression more in females than in males. Under moderate DR, ATG9A and RPS6K1 expressions were increased more in males than in females. Like females, body mass in males was correlated positively with mTOR and IGF1, but negatively with ATG9A and RS6K1 expressions. Our findings highlight that sexes may cope with nutritional deficits differently and emphasise the importance of considering sexual differences in studies of dietary restriction.

Male Japanese quails (Coturnix japonica) have been found to exhibit a three-phase metabolic change when subjected to prolonged fasting, during which basal thermogenesis is significantly reduced. A study had shown that there is a significant difference in the body temperature between male and female Japanese quails. However, whether female Japanese quails also show the same characteristic three-phase metabolic change during prolonged fasting and the underlying thermogenesis mechanisms associated with such changes are still unclear. In this study, female Japanese quails were subjected to prolonged starvation, and the body mass, basal metabolic rate (BMR), body temperature, mass of tissues and organs, body fat content, the state-4 respiration (S4R) and cytochrome c oxidase (CCO) activity in the muscle and liver of these birds were measured to determine the status of metabolic changes triggered by the starvation. In addition, the levels of glucose, triglyceride (TG) and uric acid, and thyroid hormones (T3 and T4) in the serum and the mRNA levels of myostatin (MSTN) and avian uncoupling protein (av-UCP) in the muscle were also measured. The results revealed the existence of a three-phase stage similar to that found in male Japanese quails undergoing prolonged starvation. Fasting resulted in significantly lower body mass, BMR, body temperature, tissues masses and most organs masses, as well as S4R and CCO activity in the muscle and liver. The mRNA level of av-UCP decreased during fasting, while that of MSTN increased but only during Phase I and II and decreased significantly during Phase III. Fasting also significantly lowered the T3 level and the ratio of T3/T4 in the serum. These results indicated that female Japanese quails showed an adaptive response in basal thermogenesis at multiple hierarchical levels, from organismal to biochemical, enzyme and cellular level, gene and endocrine levels and this integrated adjustment could be a part of the adaptation used by female quails to survive long-term fasting.

PROBLEM: Reproductive performance of animals gets affected by nutritional restrictions which act as potential stressors leading to hormonal imbalance and testicular inflammation, the major causes of infertility. Withania somnifera (WS), well-known traditional medicinal plant, has been used as antistress and infertility treatment. Therefore, the present study looks into the ameliorative effects of WS on the reproductive and immune system of male Coturnix coturnix japonica in stressed conditions like water and food restriction focussing on the modulation in estrogen receptor alpha (ERα). METHOD OF STUDY: Biochemical estimations for oxidative stress, histological alterations, immuno-fluorescent localization of ERα, interleukin (IL)-1ß, IL-4, and interferon gamma (IFN-γ) in testicular cells were performed. RESULTS: Nutritional restriction declines endogenous estradiol, ERα in testicular cells while it elevates corticosterone leading to oxidative stress in testis thereby reducing fertility by decrease in sperm. Results indicate significant reversal in all the parameters after the administration of WS by improving testicular cell morphology, increased superoxide and catalase activity thus reducing oxidative stress. WS increases spermatogenesis and enhances expression of ERα in testicular cells in quail. Further, WS increases IL-4, decreases IL-1ß and IFN-γ expression in testis, thereby improving immune profile contrary to stressed conditions. CONCLUSION: WS stimulates HPG-axis even after stress resulting in increased endogenous estradiol which stimulates the expression of ERα in testis; increases sperm count and immunity thereby improving the reproductive performance. WS may be the best therapy against nutritional-restriction stress induced reproductive toxicity by reducing oxidative stress mediated inflammatory response via increased testicular expression of ERα in quail.

The present study explored the neurotoxic impacts of lead (Pb) and the potential alleviating effect of Yucca schidigera extract (YSE) in Japanese quails. About 360 adult Japanese quails (8 weeks old) were used. Quails were randomly distributed to six groups with 4 replicates each: the control group (fed basal diet, BD), the BD + YSE1 and BD + YSE2 groups (BD + 100 and 200 mg/kg diet of YSE, respectively), the Pb group (BD + 100 mg/kg Pb), and the Pb + YSE1 and Pb + YSE2 groups (BD + Pb + 100 and 200 mg/kg YSE, respectively). This feeding trial lasted for 8 weeks. The exposure to Pb in the diet induced oxidative damage stress in the brain of exposed quails reflected by the significant increase in the oxidative markers including malonaldehyde (MDA) and protein carbonyl (PC) and the significant reduction in the activities of antioxidants including catalase (CAT), superoxide dismutase (SOD), and the reduced glutathione (GSH). Brain neurochemistry and enzyme activities were also altered following Pb exposure. Pb significantly reduced serotonin, dopamine, norepinephrine, GABA, Ach, and Na + /K + -ATPase activities. Pb dietary intoxication markedly increased brain inflammatory biomarkers, including tumor necrosis factor (TNF-α), myeloperoxidase, and nitric oxide. Peripherally, Pb toxicity decreased the amino acid neurotransmitters (glutamic acid, glycine, and aspartic acid) in the serum of birds. At the transcriptomic level, Pb exposure upregulated the transcription patterns of CASP3, TNF-α, HSP70, and IL-1ß. The single effect of YSE maintained that all the assessed parameters were not changed compared to the control. Interestingly, the YSE co-supplementation with Pb alleviated the Pb-induced neuro-oxidative damages by lowering the lipid, protein, and DNA damage, and the inflammatory biomarkers.

The current study was performed to determine the effect of dietary vitamin E, sesamin and thymoquinone bioactive lignans derived from sesame and black seed on immunological response, intestinal traits and Mucin2 gene expression in broiler quails. Three hundred and fifty (one days-old) quails were allotted to seven dietary treatments with five replicates as an experimental randomized design study. Treatments were basal diet as a control, control +100 and +200 mg of vitamin E, sesamin and thymoquinone per each kg of diet respectively. At 35 d of age, two quails from each pen were chosen, weighted, slaughtered, eviscerated and lymphoid organ relative weights were measured. Anti-body titers against Newcastle disease (ND), Sheep red blood cell (SRBC), and infectious bronchitis virus (IBV) and Avian influenza (AI) vaccination were determined. The serum activities of alkaline phosphatase (ALP), alanine aminotransferase (ALT), aspartate aminotransferase (AST) and serum antioxidant activates such as superoxide dismutase (SOD),glutathione peroxidase(GPX), catalase (CAT) and total antioxidant capacity (TAC) were examined. The cell mediated immunity by dinitrochlorobenzene (DNCB) and phytohemagglutinin (PHA) challenges were assessed. The microflora populations of ileum, morphological traits of jejunum and mucin2 gene expression were analyzed. Data showed that the lymphoid organ (thymus, spleen and Bursa) relative weights and antibody titer against HI, AI, SRBC and IB vaccination were increased compared to the control (p ≤ 0.05). Serum activities of ALP, ALT and AST were decreased under influences of dietary treatments (p ≤ 0.05). The serum antioxidant activates of GPX,SOD,CAT and TAC were increased and Increasing in mean skin thickness after DNCB challenge and decrease wing web swelling response to PHA mitojen injection were observed (p ≤ 0.05). Salmonella enterica, E-coli and Coliforms colonies were decrease and Lactobacillus colonies increased instead (p ≤ 0.05). The villus height and surface, crypt depth and goblet cells density were increased compared to the control (p ≤ 0.05). The expression of MUC2 gene increased under influnces of vitamin E, sesamin and thymoquinone supplemented diets (p ≤ 0.05).

In ovo exposure to o,p'-dichloro-diphenyl-trichloroethane (o,p'-DDT) impairs reproduction by inducing malformation of the reproductive organs in birds, although the mechanism remains unclear. Here, we examined the effects of o,p'-DDT on the development of the reproductive organs, the expression of genes controlling sexual differentiation, and the plasma concentrations of testosterone and estradiol in Japanese quail embryos. o,p'-DDT-containing sesame oil was injected into the yolk sac on Embryonic Day (E) 3 at a dose of 500, 2,000, or 8,000 µg per egg. On E15, the reproductive organs were observed; the gonads and Müllerian ducts (MDs) were sampled to measure the mRNA of steroidogenic enzymes, sex steroid receptors, anti-Müllerian hormone (AMH), and AMH receptor 2 (AMHR2); blood samples were collected to assay plasma testosterone and estradiol levels; and the gonads were used for histological analysis. o,p'-DDT dose-dependently increased the prevalence of hypertrophic MDs in females and residual MDs in males. In female MDs, o,p'-DDT dose-dependently decreased estrogen receptor (ER) α, ERß, and AMHR2 mRNA expression. o,p'-DDT dose-dependently induced left-biased asymmetry of testis size, and ovary-like tissue was found in the left testis after exposure to 8,000 µg per egg o,p'-DDT, although asymmetric gene expression did not occur. o,p'-DDT did not affect ovarian tissue but did decrease 17α-hydroxylase/C17-20 lyase mRNA expression and dose-dependently increased ERß mRNA expression. o,p'-DDT decreased plasma testosterone concentrations in females. These findings suggest that o,p'-DDT induces hypertrophy of the MDs and ovarian tissue formation in the left testis. Abnormal MD development may be linked to altered gene expression for sensing estrogens and AMH signals.

Due to the wide use of atrazine (ATR), the concern has increased regarding the negative impact of ATR on reproduction. Nevertheless, the reproductive effects caused by different exposure concentrations and the severity of toxic damage are poorly understood. In organisms, ATR is metabolized and degraded through phase II enzyme systems, and changes in cytochrome P450 (CYP) enzymes may have a regulatory role in the harm of ATR. However, less information is available on the induction of CYPs by ATR in avian organisms, and even less on its effects on the testis. Birds are exposed to ATR mainly through food residues and contaminated water, the purpose of this study was to examine reproductive toxicity by different exposure concentrations and elaborate metabolic disorders caused by ATR in European quail (Coturnix coturnix). In this study, the quail were given ATR at 50 mg/kg, 250 mg/kg and 500 mg/kg by oral gavage for 45 days, and the testicular weight coefficients, histopathology and ultrastructure of testes, primary biochemical functions, sex steroid hormones, critical protein levels in the testosterone synthesis pathway, the expression of genes involved CYPs, gonad axis and nuclear receptors expression were investigated. Altogether, testicular coefficient decreased significantly in the high-dose group (1.22%) compared with the control group (3.03%) after 45 days of ATR exposure, and ATR is a potent CYP disruptor that acts through the NXRs and steroid receptor subfamily (APND, CAR, ERND and ERα) without a dose-dependent manner. Notably, ATR interfered with the homeostasis of hormones by triggering the expression of hormones on the gonad axis (LH and E2). These results suggest that exposure to ATR can cause testicular toxicity involving accommodative disorder of phase II enzyme and testosterone synthesis in European quail.

One of the most intriguing areas of research and innovation in the animal production and food sector recently has been designed-enriched products. These items are regarded as functional foods because they feature components that have advantageous physiological impacts on human health. In the production of poultry, designed eggs constitute a significant category of functional foods. The present study hypothesized that adding different kinds of oils to quail diets will help produce designer eggs rich in omega-3 and 6 fatty acids in addition to enhancing productive performance. So, this study examined how linseed (flaxseed) and canola oils with various levels can affect lipid metabolism, immune function, and the amount of n-3 polyunsaturated fatty acids (n-3 PUFA) in Japanese quail eggs. This work was conducted using 3 different vegetable oils (sunflower, linseed, and canola oils) and 3 different antioxidant supplements (0, 250 mg vitamin E/kg feed, and 1,000 mg ginger/kg feed) in a 3 × 3 factorial experiment. When linseed or canola oil was added to the diet, the number of fatty acids in the egg yolks of Japanese quail layers fell by (12.7 and 18.9%) and (41.4 and 24.6%), respectively. The amounts of saturated and monounsaturated fatty acids in total eggs fell by 21.9 and 14.6% and 24.5 and 15.8%, respectively, at 20 wk of age. However, when linseed and canola oil were added to the diet, the sum n-3 PUFA content in the egg yolk of Japanese quail-laying hens was noticeably raised at 15 and 20 wk of age. At 15 and 20 wk of age, the same groups' total n-6 PUFA content considerably increased compared to the group that did not receive flaxseed. In conclusion, during the laying period of Japanese quail, linseed oil, canola oil, vitamin E, or ginger positively affected productivity, blood hematology, constituents, resistance, lipid digestion system, and antioxidative properties in serum and egg yolk.

In some areas of the world, climate-controlled poultry houses are not possible; thus, likely resulting in lower production measurements and poorer quality poultry products due to lipid oxidation during heat stress. In Japanese quail, heat stress can occur starting at 30˚C; however, as climate change becomes more severe, temperatures above 30˚C may become more frequent. Endogenous antioxidant enzymes such as superoxide dismutase (SOD) and catalase (CAT) can prevent further oxidation. The goal of this study was to determine if 10 generations of selection for low feed conversion ratio (FCR) in Japanese quail at 31.1˚C resulted in lower lipid oxidation and more antioxidant activity. The experimental design for adult tissues was 4 treatments × 2 sexes × 4 tissue types and for egg yolks was 4 treatments × 3 wk of lay with varying storage conditions. Lipid oxidation was determined in brain, liver, kidney, thigh, and yolk. SOD and CAT activities were determined in brain, liver, kidney, and thigh. ANOVA indicated significance at P ≤ 0.05. Results suggested that heat stress at 31.1˚C and 10 generations of selection for low FCR did not significantly affect lipid oxidation and antioxidant enzyme activities across all tissues. Tissue differences occurred in lipid oxidation and antioxidant enzyme activity. Brain had the most oxidation, followed by liver > kidney > thigh (P < 0.0004). Kidneys had significantly more CAT activity than brain, liver, and thigh. Brain and thigh had similar CAT activities. Thus, poultry products from quail raised at this temperature may have similar quality to those that are raised within their thermoneutral zone (18 to 30˚C). Future directions could include comparisons within the thermoneutral zone and incrementally higher temperatures to 1) to pinpoint the temperature when biochemical measurements in tissues associated with lipid oxidation begin to occur, 2) determine when total antioxidant capacity and lipid oxidation are significantly higher, and 3) ascertain SOD and CAT activity in day-of-lay yolks of eggs for future production to properly administer heat stress mitigation strategies.

Light has a very important function in the regulation of the normal physiology including the neuroendocrine system, biological rhythms, cognitive behavior, etc. The variation in photoperiod acts as a stressor due to imbalance in endogenous hormones. Estrogen and its receptors ER alpha and beta play a vital role in the control of stress response in birds. The study investigates the estrogenic effects of a well-known medicinal plant Withania somnifera (WS), mediated by estrogen receptor alpha (ERα) in the hypothalamic pre-optic area (POA) and paraventricular nuclei (PVN). Further the study elucidates its anti-oxidants and anti-apoptotic activities in the brain of Japanese quail. To validate this hypothesis, mature male quails were exposed to long day length for 3 months and then transferred to intermediate day length to become photorefractory (PR) while controls were still continued under long daylength. Supplementation of WS root extract in PR quail increases plasma estrogen and lowers corticosterone. Further, in PR quail the variation in light downregulates immunoreactivity of ERα, oxidative stress and antioxidant enzyme activities i.e. superoxide dismutase and catalase in the brain. Neuronal apoptosis was observed in the POA and PVN of PR quail as indicated by the abundant expression of Caspase-3 and p53 which reduces after the administration of WS root extract. The neuronal population also found to decrease in PR although it increased in WS administered quails. Further, the study concluded that change in photoperiod from 3 months exposure of 16L: 8D to 13.5L: 10.5D directly activates neuronal apoptosis via expression of Caspase3 and p53 expression in the brain and increases neuronal and gonadal oxidative stress while WS root extract reverses them via enhanced estrogen and its receptor ERα expression in the hypothalamic pre-optic and PVN area of Japanese quail.

Glucagon-like peptide 1 (GLP-1) elicits a potent reduction in food intake, although the central mechanism mediating this appetite-suppressive effect is not fully understood in all species. To begin to elucidate the molecular mechanisms in quail, we administered GLP-1 via intracerebroventricular (ICV) injection to 7-day-old Japanese quail (Coturnix japonica) and determined effects on food and water intake, behavior, and brain nucleus activation. We observed a reduction in food and water intake, with the lowest effective dose being 0.01 nmol. Quail injected with GLP-1 displayed fewer steps, feeding pecks, exploratory pecks, and jumps, while time spent sitting increased. We quantified c-Fos immunoreactivity at 60 min post-injection in hypothalamic and brainstem nuclei that mediate food intake and determined that the hypothalamic paraventricular nucleus (PVN), and nucleus of the solitary tract and area postrema of the brainstem were activated in response to GLP-1. In conclusion, these results suggest that GLP-1 induces anorexigenic effects that are likely mediated at the level of the PVN and brainstem.

Birds are sensitive to environmental pollution and lead (Pb) contamination could negatively affect nearly all avian organs and systems including kidney of excretive system. Thereby, we used a biological model species-Japanese quail (Coturnix japonica) to examine the nephrotoxic effects of Pb exposure and possible toxic mechanism of Pb on birds. Quail chicks of 7-day-old were exposed to 50 ppm Pb of low dose and high dose of 500 ppm and 1000 ppm Pb in drinking water for five weeks. The results showed that Pb exposure induced kidney weight increase while body weight and length reduction. The increase of uric acid (UA), creatinine (CREA) and cystatin c (Cys C) in the plasma suggested renal dysfunction. Moreover, both microstructural and ultrastructural changes demonstrated obvious kidney damages. In particular, renal tubule epithelial cells and glomeruli swelling indicated renal inflammation. Furthermore, changes in the content and activity of oxidative stress markers suggested that Pb caused excessive oxidative stress in the kidney. Pb exposure also induced abnormal apoptosis in the kidney. In addition, RNA sequencing (RNA-Seq) analysis revealed that Pb disturbed molecular pathways and signaling related with renal function. Especially, Pb exposure resulted in an increase in renal uric acid synthesis by disrupting purine metabolism. Pb caused apoptotic increment by inhibiting the phosphatidylinositol-3-kinase (PI3K)/RAC-alpha serine/threonine-protein kinase (AKT) pathway and induced aggravated inflammation by activating Nuclear Factor kappa B (NF-κB) signaling pathway. The study implied that Pb caused nephrotoxicity through structural damages, uric acid metabolism disorder, oxidation imbalance, apoptosis and inflammatory pathway activation.

Japanese quails reared under high stocking density (SD) were evaluated for the effects of grape seed powder (GSP) and meal (GSM) supplementation on performance, blood biochemistry, thigh and breast muscle fatty acids, antioxidant status, and HSP70 gene expression. We randomly assigned 288 (15-day-old) quail chicks to six treatment groups in a factorial design (2 × 3) with four replicates, involving two density levels [160 cm2/bird (LD) and 80 cm2/bird (HD)] and three feed forms (FFs) [no supplementation, grape seed powder (3% GSP), grape seed meal (3% GSM)]. SD had a significant effect on live weight, but not on weekly feed intake, daily weight gain, and feed conversion ratio. Serum creatinine and aspartate aminotransferase levels were significantly affected by FF and SD × FF (p < 0.05). A high SD reduced the n-3/n-6 ratio of breast muscle and a significant interaction was found between FF (p < 0.001). The SD × FF interaction reduced the Σn-6 ratio in HDM's thigh muscle, whereas in LDM, the ratio increased (p < 0.01). At high SD, neither GSP nor GSM reduced biological markers of oxidative stress (p > 0.05). Compared to GSP, GSM had higher efficacy at reducing HSP70 levels related to high SD levels. Despite this, at high SD, a diet containing 3% of GSP and GSM was not effective in overcoming oxidative stress. Therefore, more studies using different doses of GSM and GSP in quail diets would be beneficial.

The current study was conducted to study the impact of low-CP protein diets with different sources of zinc (zinc oxide-ZnO and zinc methionine-Zn-Met) on performance, lipid profile, immune and antioxidant parameters of quail breeders. A total number of 216 Japanese quail at 8 weeks of age were randomly distributed into nine (3 × 3) groups, with four replicates, each of six quails (four females and two males). Quails were fed three levels of CP (16, 18, and 20%) and three sources of Zn (0, 0.1 g ZnO, and 0.1 g Zn-Met/kg diets). No significant differences in daily feed intake (FI), feed conversion ratio (FCR), and egg mass are due to the main effect of protein levels or zinc sources at all ages. Dietary CP (16 and 20%) in growing quail diets increased SOD and GSH, while IgG levels were improved by 18%. Finally, it can be recommended that 16% CP and zinc source (Zn-Met or ZnO) can maintain egg mass, as well as lipid profile, immunological and antioxidant parameters in Japanese quails breeders. This level (16% CP) is enough during the production stage and a valuable way for mitigating ammonia pollution and reducing feeding costs.

The lead (Pb) contamination is considered a lethal threat to birds. However, Pb-induced hepatotoxicology especially its impacts on metabolic processes in the liver of birds is not yet fully understood. Therefore, we tried to determine the toxicological effects of Pb exposure on hepatic carbohydrate and lipid metabolism via Phosphatidylinositol 3-kinase (PI3K)/protein kinase B (Akt) pathway by using an animal model- Japanese quail (Coturnix japonica). One-week old female Japanese quails were randomly allocated into four groups and fed with 0, 50 ppm, 500 ppm and 1000 ppm Pb drinking water respectively for 49 days. The results showed that Pb accumulated in the liver as a dose-dependent manner. Exposure to high dose of Pb (500 and 1000 ppm Pb) led to severe histopathological damages characterized by irregularity and dilation of liver sinusoids, hepatic lipid vacuolization and hepatocellular cytoplasm hyalinization. Meanwhile, Pb exposure caused glycogen increase and lipid droplets decrease in the liver. Pb exposure was also attributable to a decreased triglyceride level in the plasma. In addition, the transcriptional levels of PI3K and Akt in the liver were downregulated by Pb exposure. Subsequently, the mRNA expressions of genes related with glycometabolism in the liver were remarkably altered and the mRNA levels of genes involved in fat synthesis and oxidation in the liver were also markedly changed. it seems that Pb could lead to liver metabolic disorder through structural damages and PI3K/Akt signaling pathway disruption.

Generation of egg yolk by birds requires the synthesis and deposition of large amounts of protein and lipid, and is often accompanied by the incorporation of additional physiological mediators. While there has been much work examining the relative quantities of yolk components, as well as potential adaptive patterns of their allocation, we still do not have a full understanding of what controls yolk formation and composition. Once ovarian follicles are recruited into the preovulatory hierarchy, the yolk is deposited in concentric rings, with one ring deposited per day. Previous studies have shown that there is substantial interspecific and intraspecific variation in the number of rings in yolks, and thus the number of days it took those yolks to grow. We hypothesized that the ability to grow follicles to maturity quickly is limited by the availability of materials to make yolk precursors in the female, either in body reserves or in dietary access. To test this, we supplemented the diets of Japanese quail with hard-boiled chicken yolk and examined the influences of treatment and female body condition on follicle growth rates. Contrary to predictions, females with higher body condition indices produced yolks that grew more slowly, and yolks from supplemented birds grew more slowly than controls. These results indicate that females can modulate the rate of yolk incorporation into developing follicles, and that an energy balance that is too high may not be optimal for the fast growth of developing ovarian follicles.

Lead (Pb) becomes a global public health concern for its high toxicology. Birds are sensitive to environmental pollution and Pb contamination exerts multiple negative influences on bird life. Pb also impacts on avian reproductive system. Thus, in this study, we attempted to determine toxicological effects and possible mechanistic pathways of Pb on avian testicular development by using the model species-Japanese quail (Coturnix japonica). Male quail chicks of 1-week-old were exposed to 0, 50, 500, and 1000 ppm Pb concentrations in drinking water for 5 weeks when reaching sexual maturation. The results showed that high Pb doses (500 and 1000 ppm) induced testis atrophy and cloacal gland shrinkage. Microstructural damages of both hypothalamus and testis indicated the disruption of the hypothalamus-pituitary-gonadal (HPG) axis by Pb exposure. The decrease of gonadotropin-releasing hormone (GnRH), luteinizing hormone (LH) and follicle-stimulating hormone (FSH) and testosterone (T) may also imply HPG axis disruption. Moreover, excess testicular oxidative damages featured by increasing reactive oxygen species (ROS) and malondialdehyde (MDA) and decreasing catalase (CAT), glutathione (GSH), superoxide dismutase (SOD), glutathione-S-transferase (GST), and total antioxidant capacity (T-AOC) indicated increasing risks of reproductive dysfunction by Pb. Furthermore, increasing apoptosis and upregulation of gene expression associated with cell death suggested testicular abnormal development. In addition, molecular signaling involved with steroidogenesis in the testis was disturbed by Pb treatment. The study showed that Pb could impair testicular development and reproductive function by morphological and histological injury, hormone suppression, oxidative stress, cell death, and HPG axis disruption.

Two experiments were conducted to measure the apparent and standardized ileal digestibilities (AID and SID) of amino acid (AA) of corn, wheat, soybean meal (SBM), and corn gluten meal (CGM) in growing Japanese quail from 14 to 18 (Exp. 1) and 28 to 32 (Exp. 2) d of age. The basal endogenous losses of amino acids were measured by the use of N-free diet. The birds were fed on standard diet before the use of experimental diets. The experimental diets (four ingredients) and N-free diet were randomly assigned to 5 replicate pens (30 birds per pen) and fed for 5 consecutive days. The ileal digesta were collected on d 18 and 32 for the Exp. 1 and Exp. 2, respectively. AID of lysine (Lys) in corn (P = 0.047), SBM (P < 0.001), and CGM (P < 0.001); AID of threonine (Thr) in corn (P < 0.001), SBM (P < 0.001), and CGM (P = 0.075); and AID of isoleucine (Ile) in wheat (P < 0.001), SBM (P = 0.002), and CGM (P < 0.001) were increased as the birds aged. However, AID of methionine (Met) in corn (P < 0.001) and CGM (P < 0.001), AID of arginine (Arg; P < 0.001) and valine (Val; P < 0.001) in CGM were lower in younger quails. Among indispensable amino acids, the basal endogenous losses of Thr, Val, and Arg decreased by age (P < 0.001). The average of SID of Lys, Ile, Met, Val, Thr, Arg, leucine (Leu), and histidine (His) for corn, wheat, SBM, and CGM were estimated as 83, ND, 89.4, 89.4, 92.1, 90.2, 91.9, and 90.8%; 92.7, ND, 89.1, 93.9, 87.4, 90.2, 89.8, and 88.1%; 90.3, 91.8, 94.3, 90.4, 86.5, 94.0, 84.3, and 95.0%; 82.6, ND, 74.1, 79.6, 84.4, 90.6, 85.2, and 82.4%, respectively. Based on the present study, the AID and SID coefficients of indispensable AA should be adjusted for age classes in Japanese quail during the growing period.

As a plasticizer, di-2-ethylhexyl phthalate (DEHP) has been listed as a potential endocrine disruptor by The World Health Organization. The toxicity of DEHP has been widely studied, but its toxicity on the digestive tract of birds has not been clarified. Female quail were treated by gavage with DEHP (250, 500, 750 mg/kg), with the blank and vehicle control groups reserved. The result showed that DEHP raised the damage severity grade, and decreased the ratio of villus length to crypt depth. The content and activity of cytochrome P450 system (CYP450s) were increased by DEHP. DEHP interfered with the transcription of nuclear xenobiotic receptors (NXRs), CYP isoforms, and the nuclear factor-E2-related factor 2 (Nrf2) signaling pathway. This study revealed DEHP could cause the imbalance in CYP450s mediated by NXRs, and then promote Nrf2 mediated antioxidant defense. This study provided new evidence about the mechanisms of DEHP-induced toxic effects on digestive tract.

A series of experiments was designed and conducted to determine the apparent and standardized ileal phosphorus digestibility (i.e., AIDP and SIDP, respectively) of some grains and protein meals in Japanese quail at different age classes during the growing period from the hatch to 35 d of age. Experimental diets included a PFD, to measure basal endogenous P losses (EPL), corn, wheat, soybean meal (SBM), and corn gluten meal (CGM), so as to be each ingredient the sole source of P, were developed and fed to five experimental groups with 5 replicates of 30 chicks each. Titanium dioxide as an indigestible marker was added to the diets at the rate of 5 g/kg of diet. The EPL in birds fed on PFD was estimated at 201 mg/kg dry matter intake (DMI) and quail chicks fed on PFD exhibited the lowest performance compared to chicks received dietary P regardless of P sources (P = 0.001). The estimated coefficients of AIPD (P = 0.001) | SIDP (P = 0.004) for CGM, corn, SBM, and wheat were 49.2 | 51.9%, 38.8 | 44.9%, 41.4 | 45.9%, and 33.2 | 40.1%, respectively. The mean differences between AIPD and SIPD coefficients with each ingredients including CGM (P = 0.245), corn (P = 0.169), and SBM (P = 0.169) were not statistically significant, while the comparison of those estimations for wheat (P = 0.022) showed significant differences. The present work showed that the use of direct method could successfully estimate the coefficients of P digestibility in growing quail for both the cereals and protein meals that were studied. The high relative contribution of endogenous P in young quail fed on wheat during the first 2 wk posthatch makes it inevitable to correct AIDP for EPL and evoke the implementation of SIDP coefficients in the feed formulation matrix.
